RESUMO

Cytokinin (CK) is a plant hormone that plays crucial roles in regulating plant growth and development. CK-deficient plants are widely used as model systems for investigating the numerous physiological roles of CK. Since it was previously shown that transgenic or mutant CK-deficient Arabidopsis and Centaurium plants show superior tolerance to salinity, we examined the tolerance of three CK-deficient potato lines overexpressing the Arabidopsis thaliana CYTOKININ OXIDASE/DEHYDROGENASE2 (AtCKX2) gene to 50 mM, 100 mM, 150 mM, and 200 mM NaCl applied in vitro. Quantification of visible salinity injury, rooting and acclimatization efficiency, shoot growth, water saturation deficit, and chlorophyll content confirmed that the CK-deficient potato plants were more tolerant to low (50 mM) and moderate (100 mM) NaCl concentrations, but exhibited increased sensitivity to severe salinity stress (150 and 200 mM NaCl) compared to non-transformed control plants. These findings were corroborated by the data distribution patterns according to principal component analysis. Quantification of the activity of superoxide dismutases, peroxidases, and catalases revealed an impaired ability of AtCKX2-transgenic lines to upregulate the activity of antioxidant enzymes in response to salinity, which might contribute to the enhanced sensitivity of these potato lines to severe salt stress. Our results add complexity to the existing knowledge on the regulation of salinity tolerance by CK, as we show for the first time that CK-deficient plants can exhibit reduced rather than increased tolerance to severe salt stress.

The present study evaluated over two years the seasonal dynamics of Rhipicephalus microplus in a Cerrado biome of midwestern Brazil (Goiânia, Goiás) and correlated the current climatic conditions (environmental temperature, insolation, rainfall, relative humitidy and saturation deficit of the atmosphere) of this site with 30 years ago. In addition, the data collected in the present research were compared with climatic conditions data and R. microplus population dynamics conducted by our group in other regions (Formiga, Minas Gerais and Jaboticabal, São Paulo) within the same biome. For the parasitic phase dynamics, R. microplus females (4.5-8.0 mm) counts were performed on tick natural infested cows kept in Panicum maximum paddocks. To verify the larvae dynamics on pasture the flannel dragg technique on natural infested pasture was performed, and the height of the grass was measured. Five peaks of engorged R. microplus females on animals and larvae infestations on pastures were observed. Being three and two peaks during the rainy and dry season of the year, respectively. Insolation (r = 0.8758; P = 0.00009; R²â€¯= 0.7670), rainfall (r = -0.8572; P = 0.0002; R²â€¯= 0.7348), maximum environmental temperature in summer (r = 0.9999; P < 0.0087; R²â€¯= 0.9988) and the saturation deficit of the atmosphere in autumn (r = -0.9789; P = 0.0211; R²â€¯= 0.9582) influenced the seasonal dynamics of R. microplus larvae on pastures. While the forage height did not influence the number of larvae found on pasture (r = 0.1545; P = 0.7682; R²â€¯= 0.0239). The comparison between the climatic data of the last 30 years with the current climatological data (2012 to 2019) in the state of Goiás, and the comparison with R. microplus population dynamics from other two locations in the same biome indicated that the increase of the environmental temperature due to the global warming possibly is a determining factor which increase the number of one or up two tick annual peaks. In addition, this climatic variable was responsible for increase the population density of ticks on pastures and animals observed in July/winter of the present study, in comparison to other regions with the same biome, as discussed in this work.

Dermacentor reticulatus (Ixodida: Ixodidae, Fabricius 1794) is one of the most widely distributed and abundant tick species in central Europe and is a vector for a range of pathogens. Nevertheless, many aspects of its ecology and distribution remain poorly understood. To quantify the seasonal abundance of this species in the U.K. and the environmental factors that determine this, weekly sampling at sites throughout Wales and southern England was undertaken for 12 months. This showed that the activity of adult D. reticulatus peaked February and March and that no individuals were collected between May and mid-October; no questing tick activity was observed when the 5-day average temperature was greater than 15 °C. A single nymph was collected by dragging, confirming speculation over the nidicolous status of larval and nymphal stadia. Laboratory analysis found that D. reticulatus were able survive cold shock and the lower lethal temperature was estimated to be between -18 and -20 °C. Habitat was significantly associated with tick activity, with higher numbers of ticks collected from low lying vegetation in marsh environments than from exposed grassland or woodland. A strong association was observed between activity and saturation deficit suggesting that the seasonal pattern of activity seen in the field, within the sites where it was abundant, is more strongly determined by temperature than humidity. Range expansion within the U.K. should be expected, bringing with it an elevated disease risk for animal and human hosts.

Gynaeseius liturivorus (Ehara) (Acari: Phytoseiidae) is a promising biological control agent for tiny arthropod pests, including Thrips tabaci Lindeman (Thysanoptera: Thripidae) known as a major pest of Welsh onion. In fields during summer, G. liturivorus is observed on soybean, but not on Welsh onion, even when numerous T. tabaci are present. To elucidate G. liturivorus's occurrence on soybean and Welsh onion in relation to relative humidity (RH), we examined its egg hatching on their seedlings under low RH conditions. Then we estimated the moisture transpiration from both plants. Egg hatching occurred only on soybean plants exhibiting greater moisture transpiration. Aiming at utilizing G. liturivorus as a biological control agent for Welsh onion production in summer, evaluation of its relative tolerance and compensation potential for drought injury is necessary. Therefore, we used five phytoseiid species including G. liturivorus and Neoseiulus californicus to estimate the RH and vapor pressure deficit (VPD) at which 50% of eggs hatch (RH50 and VPD50). Furthermore, we examined G. liturivorus and N. californicus egg hatching under different RH oscillation conditions. Results show G. liturivorus as the most drought-sensitive among the five species tested, but G. liturivorus is able to compensate for lethal low-RH effects on egg hatching in part by periodic exposure to high RH conditions, as observed for N. californicus.

The present study aimed to evaluate biological parameters of Rhipicephalus microplus in the non-parasitic phase in both field and laboratory conditions. It also aimed to assess correlations between duration (in days) of these parameters and climatic variables (humidity, rainfall, and soil temperature) and to estimate the annual number of generations of R. microplus in a tropical region. The non-parasitic phase of R. microplus in field and laboratory conditions was evaluated throughout the course of two years. A pasture was infested with engorged female of R. microplus, and biological parameters, including female pre-oviposition, female oviposition, egg mass incubation, larval pre-hatching phase, larval maturation, and larval longevity, were evaluated concomitantly with the collection of data on climatic conditions. The same parameters were also evaluated in a climatized chamber in the laboratory. The total duration of the non-parasitic phase in the field was longer in the dry season (1st and 4th life-cycle repetitions) than in the rainy season (2nd, 3rd, and 5th repetitions). Tick biological parameters for the non-parasitic phase in the laboratory were similar to those obtained in the field during the rainy season. The evaluated biological parameters were influenced mainly by environmental and ground-level temperatures, which modified egg mass incubation, larval pre-hatching, and larval longevity periods and, consequently, the total duration of the non-parasitic phase of the tick. The annual number of generations for the tick was estimated at five per year, which is alarming because it represents an increase, and so new studies into strategic control are needed.

Longitudinal studies are fundamental in the assessment of the effect of environmental factors on tick population dynamics. In this study, we use data from a 10-year study in 11 different locations in the Netherlands to gauge the effects of climatic and habitat factors on the temporal and spatial variation in questing tick activity. Marked differences in the total number of ticks were found between locations and between years. We investigated which climatic and habitat factors might explain this variation. No effects of climatic factors on the total number of ticks per year were observed, but we found a clear effect of temperature on the onset of tick activity. In addition, we found positive associations between (1) humus layer thickness and densities of all three stages, (2) moss and blackberry abundance and larval densities, and (3) blueberry abundance and densities of larva and nymphs. We conclude that climatic variables do not have a straightforward association with tick density in the Netherlands, but that winter and spring temperatures influence the onset of tick activity. Habitats with apparently similar vegetation types can still differ in tick population densities, indicating that local composition of vegetation and especially of wildlife is likely to contribute considerably to the spatial variation in tick densities.

We herein describe the ambush behavior of Amblyomma sculptum (Berlese 1888), a widespread and epidemiologically important tick in Brazil. Along two years of sampling by visual search in the Brazilian Pantanal, A. sculptum ticks were observed on the vegetation and in the leaf litter. Most of the ticks were observed between 10 and 50cm above ground level and less than five percent of the total were positioned below 10cm, indicating that they are seeking for middle or large-sized hosts. In both seasons, vapor saturation deficit was low during the morning. No significant relationship was found between questing ticks and daytime interval of observation or saturation deficit. However, questing tick numbers seem be higher in the end of the morning, when saturation deficit reaches its peak. Behavioral patterns of A. sculptum ticks observed in Pantanal underscore the occurrence of this tick and human contact at green anthropogenic sites. Considering A. sculptum questing behavior, inferences on human behavioral patterns that enhance or avoid contact with ticks are discussed.

Seasonally varying populations of ectothermic insect taxa from a given locality are expected to cope with simultaneous changes in temperature and humidity through phenotypic plasticity. Accordingly, we investigated the effect of saturation deficit on resistance to desiccation in wild-caught flies from four seasons (spring, summer, rainy and autumn) and corresponding flies reared in the laboratory under season-specific simulated temperature and humidity growth conditions. Flies raised under summer conditions showed approximately three times higher desiccation resistance and increased levels of cuticular lipids compared with flies raised in rainy season conditions. In contrast, intermediate trends were observed for water balance-related traits in flies reared under spring or autumn conditions but trait values overlapped across these two seasons. Furthermore, a threefold difference in saturation deficit (an index of evaporative water loss due to a combined thermal and humidity effect) between summer (27.5 mB) and rainy (8.5 mB) seasons was associated with twofold differences in the rate of water loss. Higher dehydration stress due to a high saturation deficit in summer is compensated by storage of higher levels of energy metabolite (trehalose) and cuticular lipids, and these traits correlated positively with desiccation resistance. In Z. indianus, the observed changes in desiccation-related traits due to plastic effects of simulated growth conditions correspond to similar changes exhibited by seasonal wild-caught flies. Our results show that developmental plastic effects under ecologically relevant thermal and humidity conditions can explain seasonal adaptations for water balance-related traits in Z. indianus and are likely to be associated with its invasive potential.

Objectives. To relate the effect of biotic and abiotic factors on Rhipicephalus (Boophilus) microplus tick loads on cows grazing either in intensive silvopastoral systems (ISS) (Lucerna) or in grass pastures associated with sugarcane plantations (La Isabela). Materials and methods . Tick counts were performed on 27 Lucerne breed animals that were in different physiological states, six of which were grazing on forage grass paddocks associated with commercial sugarcane plantations and the remaining animals grazed in an ISS based on Leucaena leucocephala and Cynodon plectostachyus. The tick counts were made every 15 days. The data of temperature, humidity, and radiation were taken from a weather station that was inside the ISS. Results . There was a weak relationship between saturation deficit and tick load (R2=0.34) and another between UV radiation and tick load (R2=0.205) for animals grazing in ISS. There were differences in tick counts when comparing animals of similar productivity from both systems evaluated: in La Isabela (sugarcane grass paddocks) average counts were 311 ticks perceptible to the touch (TPT) and in Lucerna (ISS farm) average counts were 206 TPT (p= 0.02). Additionally, there were greater tick counts in high productivity cows compared to low productivity cows. Conclusions . The abiotic and biotic factors of the ecosystem and animal productivity can affect the TPT counts. In ISS systems, tick counts can be lower than those observed in monoculture grazing systems.


RESUMEN Objetivos . Relacionar el efecto de algunos factores bioticos y abioticos sobre las cargas de la garrapata Rhipicephalus (Boophilus) microplus en hembras bovinas que pastorean en sistemas silvopastoriles intensivos (SSPi)(Lucerna) y en monocultivos asociados a cañaduzales (La Isabela). Materiales y métodos. Se realizaron conteos en 27 animales de raza Lucerna en diferentes estados fisiológicos, seis de los cuales se encontraban pastoreando en lotes de gramíneas forrajeras asociados con plantaciones de caña comerciales y los animales restantes pastoreaban en SSPi basados en Leucaena leucocephala y Cynodon plectostachyus. El conteo de garrapatas se efectuó cada 15 días. Los datos de temperatura, humedad y radiación se tomaron de una estación meteorológica que se encontraba en el interior del SSPi. Resultados . Se encontró una relación débil entre el déficit de saturación y los conteos de garrapatas (R2=0.34) y entre la radiación UV y los conteos de garrapatas (R2=0.205) para los bovinos pastoreando en SSPi. Hubo diferencia entre los conteos en animales con similar productividad en ambos sistemas evaluados; siendo el promedio total de garrapatas perceptibles al tacto (GPT) de 311 para La Isabela y de 206 GPT para Lucerna (p=0.02). Hubo mayor número de GPT en hembras con mayor productividad en comparación con las de baja productividad (p<0.05). Conclusiones . Los factores bióticos y abióticos del ecosistema pueden influir en el promedio de GPT, al igual que el nivel de productividad de los animales. En SSPi, la carga de garrapatas puede ser inferior a la de sistemas de pastoreo en monocultivo.

Two species of velvety tree ants, Liometopum luctuosum Wheeler, and Liometopum occidentale Emery (Hymenoptera: Formicidae), are commonly found in the western Unites States from Washington to southern California. L. luctuosum is restricted to coniferous forests in the mountains in the southern range, whereas L. occidentale is found in the lowlands. The survivorship of workers of both species exposed to several temperatures and relative humidity (RH) was determined. As temperature increased, survival of both species decreased. As the RH increased, survival of both species increased. However, L. luctuosum had higher overall survival in all treatment groups. The cuticular permeability (CP) and the rates of body water loss for each species were determined. Both species had similar CPs. Increased physiological tolerances of L. luctuosum may be an explanation for its broader distribution.

Aldehyde oxidase (AO; EC 1.2.3.1) catalyzes the final step of abscisic acid (ABA) biosynthesis, which is the oxidation of abscisic aldehyde (ABAld) to ABA. Gene expression analyses indicate that the stress-induced Pisum sativum PsAOγ isoform, which effectively uses ABAld as a substrate, is encoded by the PsAO3 gene. PsAO3 was heterologously expressed in Pichia pastoris and the recombinant PsAO3 protein revealed substrate preferences highly similar to the native PsAOγ protein present in the pea leaves and roots. Both proteins prefer indole-3-aldehyde and naphthaldehyde as substrates, although high activities against abscisic aldehyde and citral were also observed. The Km values of PsAO3 for naphthaldehyde and abscisic aldehyde (4.6 and 5.1 µM, respectively) were the lowest among the substrates tested. PsAO3 activity was almost completely inhibited by potassium cyanide, diphenyleneiodonium, and methanol. Rapidly imposed drought stress did not increase the level of PsAO3 mRNA or activity of any AO isoform, although an enhanced ABA accumulation and induction of PsNCED2 and -3 (9-cis-epoxycarotenoid dioxygenase; EC 1.13.11.51) expression, both in the pea roots and leaves, was observed. During a progressively induced drought, the level of PsAO3 transcript and PsAOγ activity increased significantly in the roots and leaves, whereas ABA accumulation occurred only in the leaves where it was accompanied by induction of the PsNCED3 expression. Therefore, we suppose that next to NCED, also AO (mainly PsAOγ) might be involved in regulation of the drought-induced ABA synthesis. However, while the "constitutive activity" of PsAOγ is sufficient for the fast generation of ABA under rapid drought stress, the enhanced PsAOγ activity is required for the progressive and long-term ABA accumulation in the leaves under progressive drought stress.
